چکیده

The recent work of Fishman, Wilhelmi, and Russell (1,2) has made available a crystalline growth hormone isolated from the bovine anterior pituitary gland. It was of considerable interest to determine the sedimentation behavior of this protein in the ultracentrifuge and, at the same time, to obtain the molecular weight by this method. The amorphous preparation of Li, Evans, and Simpson (3) has not been studied in the ultracentrifuge, although estimations of the molecular weight have been obtained by other methods (3-5). The lyophilized samples of the crystalline growth hormone were prepared as already described (2). The material was electrophoretically homogeneous at pH 4.0 and at pH 10.0. When the hormone was injected daily for 10 days into hypophysectomized male rats weighing 100 gm., it produced, at a daily dose of 20 Y, an average increase in body weight of 15.8 gm. and, at a daily dose of 100 Y, an increase in body weight of 25.0 gm. The corresponding increments in the width of the proximal epiphyseal cartilage of the tibia were 180 p and 207 p.

Sedimentation behavior of hypophyseal growth hormone.

In a previous publication, it was reported that solution of the hypophyseal growth hormone in acetate buffers results in an increment of its intrinsic viscosity with an accompanying increase of molecular weight (1). Later, Smith et al. (2) confirmed these observations in ultracentrifugal studies. The purpose of this present investigation has been to study the sedimentation behavior of the hormo...

The sedimentation behavior and molecular weight of pancreatic carboxypeptidase.

Previous studies of the physical properties of crystalline carb0xypeptidas.e have included investigations of the electrophoretic behavior (1, 2) and measurements of viscosity and diffusion (2). It was reported that the several times recrystallized protein migrated essentially as a homogeneous substance in the electrophoresis apparatus (2). We have investigated the sedimentation behavior of carb...

Effect of Imatinib on the Oogenesis and Pituitary -Ovary Hormonal Axis in Female Wistar Rat

Background Imatinib mesylate, a small-molecular analog of adenosine triphosphate (ATP) that potently inhibits tyrosine kinase activities of Bcr–Abl, PDGFR-β, PDGFR-α, c-Fms, Arg and c-kit, is one of the novel molecularly targeted drugs being introduced into cancer therapy. We tested the effect of imatinib on the ovarian histological structure and the concentration of estrogen and progesterone, ...

Responses of growth rates and growth hormone levels of African catfish (Clarias gariepinus) to stocking density

In order to estimate responses of growth rate and growth hormone of Clarias gariepinus to stocking density, fish (initial weight 30.71 ± 0.89 g) were stocked in 120-L tanks at densities of 35, 65, 95, and 125 kg/m3 for 60 days. On day 30 and 60, the fish growth rates were measured, pituitary growth hormone (GH) mRNA levels and serum GH concentrations were examined by real-time qPCR and ELISA te...

A solubility analysis of crystalline ox-liver catalase.

Sedimentation. This was observed in a Svedberg oilturbine ultracentrifuge by the method of Cecil & Ogston (1948). The hydrochlorides were dissolved in 0-2M-NaCI to a concentration of 1 g./100 ml. Owing to the relatively small molecular weights and high diffusion coefficients, clear sedimentation boundaries were not obtained; the sedimentation constants were therefore measured bythe method ofGut...
